Mean fluorescent intensity of intracellular Survivin in Ba/F3 cells expressing wild-type or ITD-Flt3 during G0/G1 phase following IL-3 withdrawal

Intracellular Survivin expression during G0/G1 phase of cell cycle in Ba/F3 cells expressing wild-type Flt3 or 3 different ITD-Flt3 shown in Figure 1A through D was determined. At each time point, cells were fixed with 1% paraformaldehyde and permeabilized with 0.25% Triton X-100/1% BSA/PBS and stained with anti-Survivin antibody followed by dual staining with 7-AAD plus PE-conjugated anti–rabbit IgG. Mean fluorescent intensity of Survivin was determined during G0/G1 phase by gating cells with 2N DNA stained with 7-AAD.
