Cell-surface expression of CD40 and CD20 in different B-lineage tumor cell lines

Cells were incubated with mouse anti-CD20 or anti-CD40 antibody, stained with fluorescein isothiocyanate–conjugated antimouse IgG, and analyzed by flow cytometry using Dako QIFIKIT (Dako North America). The number of binding sites/cell was calculated by comparing the MFI value of the stained cells with a standard curve obtained from MFIs of beads coated with a known amount of mouse IgG. The number of molecules/cell was calculated from the binding site values using GraphPad Prism software Version 4.03.
