Ex Vivo Expansion of Human CFU-Meg

CD34+ MNC (5 × 103) were cultured in serum-free medium in the presence of TPO + KL + IL-3 + IL-1α for 7, 10, 14, and 21 days, respectively. Cells were then withdrawn from the cultures and assayed for CFU-Meg as detailed in Materials and Methods. Control cultures were plated immediately in plasma clot cultures stimulated with the identical cytokine combination. CFU-Meg are given as mean ± SD of quadruplicate cultures from two separate studies, each performed with marrow cells obtained from a different normal donor. The numbers of megakaryocytes/colony are also indicated.
