Mixed leukocyte cultures were prepared between purified CD3CD4+ cells and mature irradiated DC generated from either autologous or allogeneic PBMC at a DC:T-cell ratio of 1:30. Culture supernatants were harvested after 5 days for cytokine measurements. T-cell proliferation was assessed by measuring3H-thymidine incorporation during the following 16 hours.
