BM cells were subjected to a stem cell purification procedure based on their expression of stem cell antigen-1 (Sca-1) and the lack of expression of cell surface antigens associated with differentiated hematopoietic cell lineages typical for B cells (CD45/B220), myelomonocytic cells (CD11b/mac-1), and T cells (CD4/L3T4, CD8/Ly-2), using MACS Sca-1 Multisort kit (see Materials and Methods). The fractions were analyzed by flow cytometry. The results represent 2 experiments in which the purity of Sca-1+Lin was 56.6% and 63.4%, compared with 1.7% and 1.4% in the unseparated fraction and 4.2% and 5.0% in the Sca-1+ cell fraction.
