Human Lin−CD34− cells were cultured on HESS-5 stromal layers for 4 days and CD34 expression was measured by an FACS analysis. The total cell number and percentage of CD34+ cells transferred into NOD/SCID mice were indicated. Engraftment was determined by the detection of human CD45+cells using FACS and of HLA-DRB1 DNA by PCR 6 weeks after transplantation.
