PB CD34⁺ cells purified immediately after density gradient separation (Inc⁻) or after incubation on a plastic support (Inc⁺) were cultured (1 × 10⁵ cells/mL) in a serum-free medium with or without cytokines. The cells were incubated with or without (control) SDF-1 (0.05 ng/mL). At various time points, cells were harvested and counted. In some experiments, the priming effect of SDF-1 was investigated by prior treatment of the cells with SDF-1 (0.05 ng/mL) in a cytokine-free culture medium (Stem α A) for 24 hours before culture.
