Myelomonocytic HL60 cells were pretreated with mAb anti-β₁ (K20) or control IgG (10 μg/mL each) for 30 minutes at 37°C. Cells were washed and added to the wells precoated with the specific β₂-integrin ligands keyhole limpet hemocyanin (KLH), fibrinogen, or human recombinant ICAM-1, or with β₁-integrin ligands fibronectin, laminin, collagen, or human recombinant VCAM-1. Adhesion was performed in the presence or absence of blocking mAbs anti-β₂ (60.3), or anti-α₄ (HP2.1) (20 μg/mL each). The assay was performed as described in “Materials and methods.” Values are displayed as mean absorbance ± SEM of at least 3 independent experiments. Brackets indicate significant differences (P < .01).
