Ploidy analysis of splenic megakaryocytes from c-mpl−/−mice and C57/B16 mice treated with a single injection of 5-FU (150 mg/kg) is shown. For each indicated time after injection, spleen cells were collected from 4 mice (5 mice for days 0 and 7) and stained sequentially with fluorescein isothiocyanate–conjugated anti-CD41 and the DNA intercalating fluorescent dye propidium iodide. Results are mean ± SD values from 2 independent experiments. Cells were analyzed by 2-color flow cytometry.
