The relative frequencies of CD34+, CD34+CD38+ and CD34+CD38− cells in bone marrow (BM) mononuclear cells (MNCs) were determined at the time of BM aspirations for the present studies. The values for normal controls (n = 10) represent the mean (SEM), analyzed in parallel.
CD34−, CD34+CD38+, and CD34+CD38− (and when indicated CD34+CD38−Thy-1+ and CD34+CD38−Thy-1−) cells were purified as described in “Patients, materials, and methods” and subsequently transferred to slides by cytospin centrifugation before being analyzed by fluorescence in situ hybridization (FISH).
Normal BM CD34+CD38+ and CD34+CD38− cells were sorted and analyzed by FISH in an identical manner and the percentage of cells with 3 signals was established.
