Primary lymphocyte reaction (MLR) cultures were established either with naive BALB/c CD4+ T cells and irradiated, T cell-depleted B6 splenic stimulators or with DO11 CD4+ T cells, ovalbumin peptide at 5.0 μg/mL, and irradiated BALB/c splenic antigen-presenting cells. BALB/c CD4+ cells that were tolerized to B6 alloantigen were added to some wells. Supernatants were harvested on days 3 and 5 of culture. Final concentration of naive BALB/c CD4+ cells was 0.5 × 106 cells per milliliter. Final concentration of DO11 CD4+ cells was 0.15 × 106 cells per milliliter. Tolerized cells were added at a ratio of 1 tolerized cell to 3 naive BALB/c CD4+ cells and 1 tolerized cell to 1 DO11 CD4+ cell. Values are in pg per milliliter.
