Phagocytosis mediated by FcγRI and FcγRIIIA cotransfected with the γ chain or chimeric FcγR/γ chain receptors

The experiments were conducted in transiently transfected COS-1 cells. Phagocytosis was determined by light microscopy (900-1000 cells examined in a random fashion) and is expressed as the phagocytic index (PI indicates the number of ingested IgG-coated red blood cells per 100 cells). The PI values of the FcγR/γ chimeras, corrected for individual FcγR expression (MFI), are presented as the % of the transfected Fcγ receptor + γ, considered as 100%. In general, the PI values of the cotransfected FcγR + γ and chimeric FcγRs were similar when MFI values were similar. For example, in a representative experiment, we observed a PI of 80 for cells expressing FcγRI + γ (MFI 44) and a PI of 75 for cells expressing FcγRI-γ-γ (MFI 44).
