HCMV genome equivalents per cell were determined by generating standard curves using HCMV genomic DNA and cellular genomic DNA. Two independent experiments are shown. The standard curve for actin detected 102 to 105 cell equivalents per reaction with correlation coefficients of 0.998 and 0.997, slopes of -3.45 and -3.44, and y intercept values of 36.69 and 36.76 for experiment 1 and 2, respectively. The standard curve for UL123 detected 101 to 106 genome equivalents per reaction with correlation coefficients of 0.999 and 0.996, slopes of -3.39 and -3.45, and y-intercept values of 36.48 and 34.52 for experiment 1 and 2, respectively.
Each cell population was isolated by fluorescence-activated cell sorting with purities greater than 97%.
