DC were enriched from the spleen, the thymus, and 6 LN (2 inguinal plus 2 brachial plus 2 axillary) as described in “Materials and methods.” Tissues were digested with collagenase, and the low-density cells were isolated by centrifugation on a Nycodenz gradient. The proportion of myeloid-related DC (CD11c++, CD8α−) and lymphoid-related DC (CD11c++, CD8α+) were analyzed by flow cytometry. The data are presented as the absolute number of DC per spleen, per thymus, or per 6 LN, from 3 to 5 experiments in which tissues from 3 to 8 mice were pooled for analysis.
