Figure 7.
VIP-KO pDCs recipients have higher levels of GM-CSF and inflammatory cytokines in serum and upregulation of inflammatory genes in T cells. (A-H) B10.BR mice were lethally irradiated and transplanted with 5 × 103 FACS-sorted HSCs and 106 T cells from C57B/6J donors along with 5 × 104 FACS-purified wild-type or VIP-KO pDCs. (A-C) Serum samples collected from recipient blood on days +3, +8, and +15 after transplant were assayed for cytokines (Luminex 38 PLEX). Four biological serum replicates were analyzed from days +3 and day +15 mice. Day +8 samples were analyzed in duplicate (2 technical replicates per mouse), with the same panel as day +15. (D) A time course for serial measurements of GM-CSF, IFN-γ, and TNF-α expression in donor T cells collected days +3 to +15 after transplant. Statistics: two-tail unpaired Student t test, *P < .05, **P < .01, ***P < .001. (E-H) Heatmap showing donor T cells RNA expression level from spleen of recipients, day +8 and day +15 after allo-HSCT. Mean log2 expression for each subset is shown. Four biological replicates per group.
