Overexpression of AKT or Bcl-2 was not able to inhibit the effects of the combination in transfected myeloma cell lines. AKT constructs for the activated form, wild-type, Bcl-2, and empty vectors were cotransfected with GFP into MM.1S cells. Cells were then treated with control, bortezomib, lonafarnib, or the combination, and were subsequently stained with APC-annexin V. (A) The fraction of annexin V-positive, GFP-positive cells treated with bortezomib and lonafarnib when transfected with constitutively active AKT, WT AKT, WT Bcl-2, or empty vector. (B) Gating strategy to isolate the fraction of GFP-positive cells that were then analyzed for annexin V staining.
