Formation of canine FVIII-neutralizing antibodies in hemophilia A mice. (A) Correlation between the overexpression of canine FVIII and subsequent presence of FVIII-neutralizing antibodies in mice in which FVIII activities were detectable before the onset of FVIII-neutralizing antibodies. The FVIII activities at weeks 2 and 6 after vector injection were determined by Coatest. The FVIII-neutralizing antibodies at weeks 2 and 6 were determined by Bethesda assay. (B) Lack of correlation between the levels of FVIII-neutralizing antibodies and transgene DNA in liver in individual mice. The copy numbers of canine FVIII DNA per diploid genome in mouse liver were quantified by Southern blotting analysis.
