Decreased expression of adhesion molecules and other target genes in MMSET knockdown and knockout cells. (A) Quantitative real-time PCR was performed on cDNA prepared from empty vector lentivirus– or MMSET shRNA lentivirus–transduced KMS-11 cells. Expression for each gene was normalized to GAPDH expression levels. Control cell gene expression was set at 100%, and the level in MMSET shRNA cells is expressed relative to controls. SD values are shown. (B) Q-PCR was performed as in panel A using cDNA from KMS-11 cells with disruption of the translocated MMSET allele (T-KO) or a random-integrant control clone.
