MSC inhibitory effect does not require CD4⁺/CD25⁺ regulatory T cells.

CD25-depleted or CD25-replete C6 splenocytes (5 × 10⁶) were stimulated in vitro with HY K^kSmcy peptide pulsed female cells in the presence or absence of MSC (10⁵). After 24 hours HY-specific IFN-γ⁺CD8⁺ T cells were enumerated. MSC inhibition on CD25⁻ responders was 50% as compared to 44% in the control CD25-replete cultures (not statistically significant).
