After transduction the cells were mixed 1:1 and in total 1 × 106 cells were injected into the thymus transplanted subcutaneously 8 weeks prior to injection of the transduced cells. Two weeks later the mice were killed, the thymus was removed, and single-cell suspensions were made by gently cutting the thymus and pressing the fragments through a stainless sieve. The cells were stained with antibodies against the NGFR, CD123, CD4, CD8, CD45RA, BDCA2, and HLA-DR.
