Bcrp-1 expression in SP- or MP-CD34^-KSL cells. (A) The expression of Bcrp1 and Gapdh was examined by RT-PCR. cDNA was synthesized from total RNA extracted from SP- or MP-CD34^-KSL cells. Normalized amounts of cDNA templates were amplified by 32, 36, or 40 cycles. (B) The expression of Bcrp-1 was examined by immunostaining with anti-Bcrp-1 antibody. SP-CD34^-KSL cells and MP-CD34^-KSL cells were stained with anti-Bcrp-1 antibody, followed by incubation with Alexa-647-conjugated anti-rabbit IgG and DAPI. At least 50 cells were analyzed with a confocal microscope. Two cells are representatively shown for each cell type. Bcrp-1 seems unevenly distributed in the cytoplasm and/or membrane.
