Figure 4. The effect of Gardos channel blockers and valinomycin on collagen-plus-thrombin–induced procoagulant response in HEPES/choline buffer. Platelets at 107 mL–1 in HEPES/choline were preincubated with inhibitors for 30 minutes followed by activation with collagen (10 μg/mL) plus thrombin (4 nM) for 10 minutes. Valinomycin was added 5 minutes prior to activation. Samples from these incubations were diluted 10-fold and subsequently analyzed for binding of FITC-conjugated annexin A5. Data are expressed as a percentage of the number of annexin-positive cells found upon activation in HEPES/choline in absence of the inhibitors. Data are mean values ± 1SD(n = 4).
