Expression of wild-type C2GnT-I in HH cells renders the cells susceptible to galectin-1 cell death. (A) HH cells were transfected with wild-type C2GnT-I. Pooled transfected cells were analyzed by staining with 1D4. Cells transfected with C2GnT-I (solid line) demonstrated increased reactivity with 1D4 compared with cells transfected with vector alone (dotted line). Marker shows 1D4+ region (R2). (B) Pooled HH cells transfected with wild-type C2GnT-I or vector alone were treated with galectin-1 for 4 hours and analyzed for binding of annexin V. C2GnT-I–transfected cells (1D4+ cells gated in R2, solid line) demonstrated increased annexin V binding after galectin-1 treatment compared with cells transfected with vector alone (dotted line). (C) Galectin-1 cell death was determined for cells transfected with vector alone versus 1D4+ cells transfected with C2GnT-I. 1D4+ cells demonstrated significantly greater cell death compared with cells transfected with vector alone (*P < .01).
