WP1130-induced apoptosis results from down-regulation of Bcr/Abl. (A) K562 cells (1 × 106) were incubated in medium with or without WP1130 (5 μM) for the indicated times, and cell-cycle distribution was determined by FACS analysis. (B). K562 cells were electroporated with either 100 nM scrambled siRNA (control) or 100 nM siRNA specifically targeting Bcr/Abl for the indicated times, and expression of Bcr/Abl, pBcr/Abl, pCrk1, and Crk1 was determined by Western blotting. The phosphorylation of CrkL was used as an indicator of Bcr/Abl activation.
