Effects of PHA-739358 on apoptosis and CrkL phosphorylation status of CD34+ cells from an IM-resistant CML patient. CD34+ cells derived from a patient with IM-resistant blast crisis were exposed to 5 μM PHA-739358 or 5 μM IM. Analysis of the cell cycle and the apoptotic fraction of propidium iodide–stained cells was assessed by flow cytometry in untreated cells serving as a control (A), cells treated for 96 hours with IM (B), and cells exposed to PHA-739358 for 72 hours (C) and 96 hours (D). Analysis of the expression of P-CrkL by intracellular flow cytometry. Strong reduction of CrkL phosphorylation was seen after exposure to PHA-739358 (72 hours and 96 hours; F and G, respectively) with no significant changes after 96 hours of IM treatment (E).
