Anti-CD3 preconditioning separated GVL from GVHD in recipients conditioned with TBI. BALB/c mice were preconditioned with anti-CD3 on day −9. The mice were conditioned with 800 rads sublethal TBI on day 0. Six hours later, the mice were injected intravenously with TCD-BM cells (5 × 106) and spleen cells (2.5 or 5 × 106) from C57BL/6 donors. There were 12 mice in each group combined from 3 replicate experiments. (A) Clinical score. (B) Survival percentage. (C,D) Liver, skin, and colon tissues from the recipients conditioned with TBI with or without anti-CD3 preconditioning were evaluated for tissue inflammation and damage 60 days after HCT. (E-G) BALB/C recipients that were preconditioned with anti-CD3 were injected intravenously with BCL1 cells transfected with luciferase (Luc+) and donor TCD-BM and spleen cells (2.5 × 106). There were 8 mice in each group combined from 2 replicate experiments. The survival percentage, representative photographs of in vivo BLI of Luc+ BCL1 cells, and the intensity (photo/second) of BLI are shown.
