EGR-1 is the direct downstream target gene of JUN. (A) Western blot analysis compared OCI-My5 (OCI), OPM2, and U266 myeloma cells when untransfected (CON), carrying empty vector (EV), or overexpressing JUN (JUNH). Results showed specificity of JUN overexpression and effects on regulation of EGR-1. β-Actin was used as a loading control. (B) Effect of nonspecific scrambled shRNA (SCR) and JUN shRNA (JUN-KD) in U266 and OCI-My5 (OCI) cells on regulation of EGR-1 expression was evaluated by Western blot.
