To generate moDCs, the plastic-adherent fraction of PBMCs from healthy donors was cultured in medium supplemented with granulocyte-macrophage colony-stimulating factor and IL-4. After 6 days of culture, cells were matured for an additional 48 hours with IL-1β, IL-6, PGE2, and TNF. Cells were harvested on days 1, 4, 5, 6, 7, and 8, and analyzed for DEC-205, CD14, and CD83 expression (A).
