Druggable genome siRNA screen to identify genes that affect bortezomib sensitivity. (A) Heat map of raw data of all 384-well plates in primary HTS. KMS11 cells were reversely transfected with 2 siRNAs per gene from druggable genome siRNA library alongside staggered negative and positive control siRNAs. After 24-hour transfection, various doses of bortezomib were added and cell viability was determined at 96 hours (72 hours after bortezomib) by CellTiter-Glo luminescence assay read. The collected data were imported to generate this heat map to overview screen results and to evaluate screen quality. (B) The representative curve-fitting result from secondary screening.
