HLA class II antibody induced TRALI in an ex vivo rat lung model. Isolated rat lungs are perfused with human neutrophils, and supernatant from human monocyte cultures were incubated with matched or unmatched human HLA class II antibodies or human control plasma as indicated. (A) No significant increase in the capillary filtration coefficient (Kfc) or lung weight occurred in ventilated and perfused rat lungs in the absence of LPS prestimulation (P > .05 for matched vs unmatched supernatant at t = 140 minutes for both parameters comparing matched and nonmatched supernatant). (B) After prestimulation of rats with LPS 2 hours before ex vivo lung perfusion, a significant increase in Kfc and lung weight is observed after 35 minutes in the presence of matched monocyte supernatant (P < .01); n = 7 for each experiment.
