Cytokine profile and transcription factor expression of regulatory DC-maintained long-lived CD4 T cells. (A) Long-lived CD4 T cells (Longeval CD4 T cells) or naive CD4 T cells were stimulated with mDCs and 200nM OVA_323-339 for various times, and the supernatants were collected for assay of cytokines by ELISA. (B) Long-lived CD4 T cells or naive CD4 T cells were cultured with mDCs in the presence of 200nM OVA_323-339 for 18 hours and assayed by intracellular cytokine staining for IL-2, IFN-γ, IL-4, IL-5, and IL-10 expression. Numbers in plots indicate the percentage of various cytokine-producing CD4⁺ T cells. (C) Various CD4⁺ T cells were detected for T-bet and GATA-3 expression by intracellular staining. CD4⁺ thymocytes were used as a positive control of GATA-3 expression. Thin solid line, background staining. Shaded histograms, specific antibodies.
