TNT003 prevents CA-mediated hRBC lysis and anaphylatoxin generation. (A) hRBCs exposed to CAD plasma sample P5 followed by 25% NHS exhibited complement-dependent hemolysis that was inhibited by TNT003, but not IC, in a concentration-dependent manner (IC₅₀ = 5.1 μg/mL; 34 nM). Supernatant from these experiments was assayed for C4a (B; IC₅₀ = 5.2 μg/mL; 35 nM), C3a (C; IC₅₀ = 5.2 μg/mL; 35 nM), and C5a (D; IC₅₀ = 4.9 μg/mL; 33 nM) by plate-based ELISA.
