DNT-like cells have reduced CD4 or CD8 coreceptor expression. (A) CCR7⁻CD45RA⁺ CD4⁺TEMRA or (B) CCR7⁻CD45RA⁺ CD8⁺TEMRA cells were gated and analyzed for their CD27 and CD28 expression. Subsequently, CD4 or CD8 expression was analyzed on CD27⁻CD28⁻TEMRA cells (gray tinted) vs CD27⁺CD28⁺TEMRA cells (black line). For CD8⁺TEMRA the CD8 expression level (CD8 MFI, mean fluorescence intensity) is compared between CD27⁻CD28⁻ and CD27⁺CD28⁺TEMRA in 8 ALPS-FAS patients vs adult controls. Because very few healthy donors had detectable CD4⁺TEMRA, only representative plots are shown.
