Platelet-dependent replication of the Dengue virus genome. Intact platelets (squares) or platelets subjected to 3 cycles of freeze-thaw at −80°C (circles) (5 × 105/mL) were incubated with DENV2 (5 × 106 virus particles/mL) for 1 hour. After washing 3 times and resuspending the platelets in serum-free M199, the pellets were cultured at 25°C or 37°C with daily medium changes. Viral genome RNA was quantified by qRT-PCR on the indicated day. Preinoculation (open symbols) is shown on day zero (n = 3, ±SD smaller than the size of symbols).
