Shed vesicles block anti-Fas–mediated CX-1 cell death. Negative and positive controls were established by treating 2 × 105 CX-1 cells with 100 ng noncytotoxic, isotype-matched antibody (IsoAb), or antihuman Fas IgM (Anti-Fas Alone), respectively. Viability of CX-1 cells was assessed in anti-Fas IgM-containing cultures prepared with 40 μg total membrane protein from vesicles shed from MIP-101 or CX-1 cells, as indicated. Note that, although Fas-mediated apoptosis is abolished in the presence of vesicles shed from MIP-101 cells (relative to anti-Fas alone, P < .01), apoptosis is unaffected (P > .10) by the addition of vesicles shed from CX-1 cells.
