Cells obtained in X-VIVO 15 medium were functional DCs. (A) DCs obtained from the same ACs cultured either in RPMI-10% FCS (solid symbols) or in X-VIVO 15 medium (open symbols) were used in MLR with 1.5 × 10⁵ allogenic highly purified CD3⁺ T cells obtained by immunomagnetic depletion. TNF-α was added (triangle) or not (square) at day 5 of DC culture. This comparison was performed with ACs from two patients and gave the same results. (B) DCs obtained after 5 days of culture in X-VIVO 15 supplemented with GM-CSF and IL-4 were incubated with FITC-dextran and analyzed as described in the legend to Fig 5.
