MPC support of human hematopoiesis in vitro.

MPCs plated at 2 × 10⁴/cm² were treated with 10 U/mL IL-1β for 24 hours and maintained with 12.5% FCS and 12.5% horse serum. Following irradiation at 2 Gy, cord blood CD34⁺ cells were plated in contact with the MPCs. The number of CFCs present in fresh CD34⁺ cells or in progeny from CD34⁺ cells cultured on MPCs was enumerated by methylcellulose assay (n = 3). As control, cells were plated on irradiated confluent AFT024 feeders.
