DC-10 produce high levels of IL-10 but low amounts of IL-12. (A) DC-10 or myDCw FACS-sorted from peripheral blood according to CD11c, CD83, and CD14 expression were activated with zymosan, poly(I:C), LPS, flagellin, and CpG. As control, sorted CD14+ cells were used. Culture supernatants were collected after 48 hours, and levels of IL-6, IL-10, IL-12p70, and TNF-α were determined by Bioplex. (B) Monocyte-derived DCs were differentiated in IL-4 and GM-CSF in the presence of IL-10 for 7 days (DC-10), or in IL-4 and GM-CSF for 7 days (iDC), or in IL-4 and GM-CSF for 5 days and activated for additional 2 days with LPS (mDC). iDCs, DC-10, and mDC were cultured in medium or activated with IFN-γ and LPS for additional 2 days and levels of IL-6, IL-10, IL-12p70, and TNF-α in culture supernatants were determined by ELISA.
