Contribution of IFN-γ to IL-12 production in BMMCs upon LPS stimulation. (A) Real-time PCR (top) and RT-PCR analysis (bottom) of ICSBP mRNA from BMMCs that were incubated with 100 U/mL IFN-γ for 16 hours after the cells were treated with 10 μg/mL control rat IgG or anti–IFN-γ antibody for 48 hours. These results are representative of 3 independent experiments that had similar results. (B) ELISA analysis of p40 production from control IgG– or anti–IFN-γ antibody–treated BMMCs. The cells were incubated with 10 μg/mL control rat IgG or anti–IFN-γ antibody for 48 hours. Then the cells were incubated with the indicated concentration of LPS for 24 hours after the cells had been preincubated in the presence or absence of 100 U/mL IFN-γ for 16 hours.
