Irradiation increases CD153 expression on myDCs and pDCs after BMT. The surface marker CD153 was measured by FACS analysis on different dendritic cell subsets in irradiated compared with nonirradiated wt Balb/c (A) or Rag2−/− γc−/− Balb/c (B) recipients. Where indicated, DCs were derived from animals that received TCD-BM and T cells as described for the C57B/6→Balb/c model. Recipient-type myeloid DCs (CD11c+CD11b+Gr-1−) or plasmacytoid DCs (CD11c+B220+CD11b−) derived from the spleen on day 3 after BMT display differential CD153 surface expression when isolated from untreated Balb/c mice or nonirradiated Rag2−/− γc−/− mice that received a transplant of TCD-BM and T cells. Grey open histogram: isotype IgG. CD153 expression on myDCs from irradiated compared with nonirradiated Balb/c mice (A) (MFI: 110 versus 24, P < .05) or Rag2−/− γc−/− mice (B) (MFI: 195 versus 42, P < .05). CD153 expression on pDCs from irradiated compared with nonirradiated Balb/c mice (A) (MFI: 182 versus 26, P < .05) or Rag2−/− γc−/− mice (B) (MFI: 137 versus 21, P < .05).
