C3aR/C5aR are required for efficient OC differentiation. (A) 2 × 106 WT,C3aR−/−,C5aR−/−, and C3aR−/−C5aR−/− BM cells were cultured in α-MEM/10% heat-inactivated media in each well of a 24-well plate together with 1 × 10−8M 1,25(OH)2 vitamin D3. OCs were stained, and TRAP-positive cells in each well were counted under a microscope. (B) WT BM cells (2 × 106) were cultured in α-MEM/10% heat-inactivated FBS media in each well of a 24-well plate together with 1 × 10−8M 1,25(OH)2 vitamin D3 in the presence of placebo (control), C3aRA, C5aRA, or C3aRA·C5aRA. OCs were stained, and TRAP-positive cells in each well were counted under a microscope. Results are representative from 2 independent experiments performed in triplicate.
