Whether the fusion protein directly binds and up-regulates expression of MPL and Bcl-xL or whether it suppresses expression of transcriptional repressors is unclear. Note that t(8;21) AML, unlike myeloproliferative neoplasms, requires THPO for enhanced JAK/STAT and PI3K/AKT signaling through wild-type MPL.
