Targets of intervention by putative small-molecule inhibitors around the cytokine receptor, JAK-STAT, MAPK, and PI3K-mTOR pathways. Such targets include the interaction between JAK2 V617F and cytokine receptors, the pseudokinase domains of JAK2 V617F, and direct inhibition of STATs and the MAPK and PI3K pathways downstream of either cytokine receptors or tyrosine kinase receptors such as the IGF1 receptor, which could itself be targeted.
