Activity and specificity of MLL-AF4 siRNAs. (A) SiRNA scan of the MLL-AF4 mRNA fusion site. MLL-AF4 mRNA levels normalized by GAPDH mRNA levels are shown. Target sites of the indicated siRNAs were moved by one single nucleotide from the AF4 part to the MLL part of the fusion site. Total RNA was isolated 24 hours after electroporation with 500 nM of siRNA and analyzed by real time RT-PCR. One of 2 experiments yielding similar results is shown. (B) Time course of MLL-AF4 depletion. Total RNA was isolated at the indicated time points after electroporation with 750 nM siRNA.
