Effect of α-amanitin on TF activity generation during HUVEC/M coculture. α-amanitin (10 μg/mL) was incubated with HUVEC or M for 2 hours. At the end of incubation, HUVEC (2 × 105/well) and M (4 × 105/well) were washed and incubated for an additional period of 3.5 hours with untreated M and HUVEC, respectively. Cells were then processed for TF activity assay, as for Fig 1. Results are expressed in percent of TF activity. A 100% value was assigned to the activity expressed by the HUVEC/M coculture.
