Forskolin (20 μmol/L) and C2-ceramide (50 μmol/L) were then added to cells where indicated. Cells treated with C2-ceramide were also incubated in the presence of IL-3. After 6 hours at 37°C, cells were isolated, lysed, and DNA fragments resolved on 2% agarose gels and visualized by ethidium bromide staining.
