The STAT5A/WKR and ▵53C mutants inhibit G-CSF–dependent granulocytic colony formation in primary murine bone marrow. Primary murine bone marrow was transduced with parental MINV retrovirus (MINV) or MINV retrovirus expressing STAT5A/WKR (MINV/WKR) or STAT5A/▵53C (MINV/▵53C) and then plated in triplicate in methylcellulose culture with G418 and exogenous G-CSF. The mean number of granulocytic colonies on day 4 to 5 per 105 cells plated is shown and is representative of three independent experiments. Error bars indicate the standard error. No colonies were observed in sham-infected cultures selected in G418 or upon omission of G-CSF from the medium.
