Subcellular fractionation of unstimulated peripheral blood eosinophils (5 × 10⁷) obtained from an asthmatic donor. Fractions were collected from a 0% to 45% linear Nycodenz gradient and analyzed for marker enzyme activities to obtain profiles of subcellular compartments. Marker assays used were eosinophil peroxidase (secretory granules), β-hexosaminidase (secretory granules and lysosomal granules), CD9 (plasma membrane), and lactate dehydrogenase (cytosol). Quantification of RANTES was performed by ELISA for each fraction and is expressed as pg/fraction.
