Day-14 clones were stimulated with PMA and ionomycin for 24 hours and stained using FITC- and PE-labeled antibodies. (A) The surface phenotype of a representative Tc1 clone (left panel) and Tc2 clone (right panel). The percentage of cells positive for a particular marker is indicated. Tc0 clones were found to be very similar to Tc2 clones in terms of surface markers (not shown). Fas L expression was measured 6 hours after stimulation (A) and 18 hours after stimulation in the presence of protein secretion inhibitor (B). Perforin was measured by intracellular staining.
