Profile of heparin-agarose chromatography of TFPIC127.
Heparin-agarose chromatography was carried out using a 0.1 to 1 mol/L NaCl gradient in 0.05 mol/L Tris-HCl, pH 7.5. Elution of TFPIC127was monitored by a modified ELISA as described in the “Materials and Methods.” Inset: Western blot analysis of TFPIC127. Purified TFPIC127 (1 μg) was run on a 10% SDS-PAGE gel and transferred to nitrocellulose membrane. The membrane was blotted with rabbit antisera containing a peptide antibody against the last 12 amino acid residues of TFPI, and developed with ECL reagents.
